O Anticorpo MCT4 (D-1) é um anticorpo MCT4 monoclonal de camundongo IgG2a κ (também designado anticorpo SLC16A3, anticorpo membro 3 da família 16 de transportadores de soluto ou anticorpo MCT3) que detecta a proteína MCT4 de origem de camundongo, rato e humana por WB, IP, IF, IHC(P) e ELISA. O anticorpo MCT4 (D-1) está disponível na forma de anticorpo anti-MCT4 não conjugado, bem como em várias formas conjugadas de anticorpo anti-MCT4, incluindo conjugados de agarose, HRP, PE, FITC e vários conjugados Alexa Fluor®. Os monocarboxilatos, como o lactato e o piruvato, desempenham uma função integral no metabolismo celular. O ácido lático é produzido em grandes quantidades como resultado da glicólise, que fornece a maior parte do ATP às células em condições fisiológicas normais. Entretanto, o acúmulo de ácido lático leva a uma diminuição do pH intracelular e à interrupção da glicólise. Para que a glicólise continue em uma taxa alta, o ácido lático deve ser transportado para fora da célula. Esse processo de transporte é realizado por uma família de transportadores de monocarboxilato (MCTs), que funcionam como simpatizantes de prótons e são estereosseletivos para o L-lactato. A família MCT consiste em pelo menos oito membros, MCT1-8, que contêm entre 10 e 12 domínios transmembrana-helicoidais (TM), com os terminais amino e carboxi localizados no citoplasma. A MCT1 é amplamente expressa e é a principal forma de MCT em células tumorais e eritrócitos. O MCT2 é altamente expresso no fígado e no testículo, enquanto o MCT3 e o MCT4 são predominantemente expressos no músculo esquelético.

I am planning to use this antibody for detecting mouse MCT4 by FCM. Can you confirm that this antibody specifically targets the cytoplasmic domain of MCT4 and is suitable for use in both mouse and human samples?

Which specific antigen retrieval do you recommand using MCT4 (D-1): sc-376140 antibody between :10 mM sodium citrate buffer, pH 6.0 (sc-294091); 50 mM glycine-HCl buffer (sc-29096), pH 3.5, and 0.01% (w/v) EDTA (sc-29092)?

Thank you for your question. For antigen retrieval in experiments with this antibody we recommend heat treatment: Heat treatment (recommended method): Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

I tried on mouse sample (4T1 cells lysate ) for WB, but doesn't work. is it possible to have the an aliquot of the discontinued MCT4 (sc-50329) that was working in my previous experiments?
