O Anticorpo Recetor de Estrogénio alfa (D-12) é um anticorpo monoclonal de cadeia leve IgG2a kappa de camundongo criado contra o mapeamento dos aminoácidos 2-185 no terminal N do Recetor de Estrogénio alfa humano (ERα). O anticorpo monoclonal ERα (D-12) detecta ERα de origem de camundongo, rato e humana por western blotting (WB), imunoprecipitação (IP), imunofluorescência (IF), imunohistoquímica em secções incluídas em parafina (IHCP) e ensaio de imunoabsorção enzimática (ELISA). A proteína ERα localiza-se principalmente no núcleo das células responsivas aos estrogénios, onde ERα funciona como um fator de transcrição ativado por fixador pertencente à superfamília dos receptores de hormonas esteróides/tiroideas. A localização nuclear do ERα é crucial porque o ERα permite a interação direta com o ADN nos elementos de resposta aos estrogénios (EREs), regulando a transcrição de genes envolvidos na proliferação celular, diferenciação e funções reprodutivas. I would like to know if this antibody recognize the whole sequence from 2- 185, or just part of the sequence within?

Thank you for your question. Estrogen Receptor alpha Antibody (D-12): sc-8005 was raised against amino acids 2-185 mapping at the N-terminus of Estrogen Receptor alpha of human origin. Its binding site is one specific point within this epitope region.

Can we use this antibody for IF and IHC staining in paraffin-embedded mouse tissues?

ER&alpha; Antibody (D-12): sc-8005 is recommended for detection of ER&alpha; of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA, so using it for IF or IHC(P) staining should work well. For published data with this antibody in murine tissues and IHC(P) please see PMID 19332863 or PMID 22992387.

Can we use this antibody in rat brain samples for W.B. Immunoflourescence in frozen sections?

Thank you for your question. This mouse monoclonal antibody, ER&alpha; (D-12): sc-8005, is suitable for Western blotting (WB) and immunoflourescence (IF) in frozen tissues of rat origin. We have not tested this antibody specifically in rat brain tissue samples. Please refer to the data on our website for our recommended positive controls to include when testing this antibody in your samples.

Can I use this antibody for immunoprecipitation of ER alpha from mouse brain?

Thank you for your question. This mouse monoclonal antibody, ER&alpha; (D-12): sc-8005, can be used for immunoprecipitation (IP) in mouse samples. We are not aware of any specific publications citing use of the antibody to IP ER alpha from brain tissue samples, but it has been successfully used for IP in both human and mouse cell lines.
