Chibby Antibody (C-10) is a mouse monoclonal IgG2b kappa light chain antibody that detects Chibby protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Chibby antibody (C-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Chibby protein, also known as Cytosolic leucine-rich protein or PIGEA-14, is a highly conserved 126 amino acid protein that plays a crucial role in inhibiting β-catenin-mediated transcriptional activation by competing with LEF-1 for binding to β-catenin. This function is vital for regulating Wnt signaling pathways, which are essential for various cellular processes, including cell proliferation and differentiation. Chibby is primarily localized in the nucleus and the Golgi apparatus within the trans-Golgi network, and acts as a homodimer. Interaction with the 14-3-3 protein leads to sequestration of Chibby in the cytoplasm, forming a stable complex with β-catenin that facilitates nuclear export of β-catenin. While Chibby is widely expressed, particularly high levels are found in skeletal muscle, heart, placenta, and kidney. Down-regulation of Chibby has been observed in thyroid and metastatic uterine tumors, indicating that the gene encoding Chibby may function as a tumor suppressor, highlighting potential importance in cancer biology.
