ZSWIM8 (zinc finger SWIM-type containing 8) encodes an E3 ubiquitin ligase component that functions as a substrate adaptor within a Cullin-RING ubiquitination complex, helping control protein turnover in the nucleus and cytoplasm. A key role of ZSWIM8 is in small-RNA quality control, where it promotes target-directed microRNA degradation (TDMD) to tune miRNA abundance and downstream post-transcriptional gene regulation. Through these mechanisms, ZSWIM8 influences pathways linked to cell-state transitions, developmental programs, and stress-responsive transcriptional networks. Dysregulated ubiquitin-mediated proteostasis and miRNA homeostasis associated with altered ZSWIM8 activity have been investigated in contexts relevant to cancer biology and neurodevelopmental phenotypes.
ZSWIM8 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ZSWIM8 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ZSWIM8 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ZSWIM8 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ZSWIM8 protein expression.
This CRISPR knockout system enables efficient generation of ZSWIM8-deficient cell models for investigation of ZSWIM8 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
