ZNF76 Antibody (F-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects ZNF76 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ZNF76 Antibody (F-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ZNF76, also known as ZNF523 or Zfp523, is a transcriptional repressor predominantly expressed in testis, where ZNF76 plays a crucial role in regulating gene expression. ZNF76 is the human homolog of Xenopus Staf protein, which is essential for transcription of genes encoding small nuclear RNA and selenocysteine tRNA. ZNF76 is localized in the nucleus, where ZNF76 exerts an inhibitory effect on p53-mediated transactivation, specifically targeting TFIID complex through interactions with both N and C termini. Post-translational modifications, particularly lysine acetylation and sumoylation, intricately regulate ZNF76 functional activity by influencing ZNF76′s ability to interact with TFIID. Acetylation by p300 leads to reduced sumoylation and weakened interaction with TFIID, while deacetylation by HDAC1 can restore ZNF76′s repressive function. Alternative splicing produces two ZNF76 isoforms, which further diversifies regulatory capabilities and interactions within transcriptional machinery.
