ZEB2 Antibody (E-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects ZEB2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-ZEB2 antibody (E-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ZEB2, also known as zinc finger E-box-binding protein 2, plays a pivotal role in regulating gene expression through interactions with transcription factors, particularly in epithelial-mesenchymal transition (EMT), which is crucial for processes such as embryonic development and tissue repair. ZEB2 is characterized by a unique structure, including a SMAD-binding domain, a homeodomain, and two clusters of zinc fingers at both N- and C-termini, allowing binding to specific DNA sequences and interaction with other proteins. This structural complexity is essential for mediating the effects of transforming growth factor-beta (TGF-β) signaling, vital for normal neural development and formation of neural crest cells. Mutations in the ZEB2 gene, located on chromosome 2q22, are associated with Hirschsprung disease, leading to severe developmental disorders such as mental retardation, delayed motor skills, and congenital heart defects, underscoring ZEB2′s significance in both health and disease.

I'm thinking about using this antibody for immunohistochemistry. Could you tell me the condition of antigen retrieval? (HIER or PIER? etc)

I'm confused, in the datasheet it mentions that the size is 157kDa, but that is not the size in the western blots shown (at least not in one of them with three different lysates). Why is this? What is then the expected size we have to look for?

Thank you for your question. According to Uniprot (http://www.uniprot.org/uniprot/O60315), the predicted molecular weight for ZEB2 is 136 kDa. On our product data sheet, we list 157 kDa as the expected molecular weight, which is also a prediction based on a peptide calculator. This is also consistent with the molecular weight that we observed when using this antibody for Western blot in human brain tissue extract. The Western blot data currently shown with three different lysates shows a signal detected between 90-132 kDa. Because we can not find any current reference supporting this molecular weight for ZEB2, we will remove this data from our website.

I am going to use this antibody on my mouse cells. Any tips for how to do this with mouse monoclonal antibodies?

We recommend using conjugated primary antibodies. This product, sc-271984, is available directly conjugated to FITC, AF488, and AF 647.

Rated 3 out of 5 by from Not good.I got the free sample of ZEB2 antibody. The major band is at around 92kd, a weak band was shown at ~200kd size.However, the predict size should be ~136kd.

Rated 1 out of 5 by from No band at appropriate positionWe bought the antibody ZEB2 (E-11) sc 271984 and since then we have performed several western blots with this antibody. However we have never seen a band at ~157kDa (the molecular weight provided in the data sheet). There are reports available for ZEB2 band at ~150kDa in the cells that we are using (dendritic cells) however we always see a band at ~75kDa. Although it is a monoclonal antibody we have not seen the band at the right position even once. I am putting up a picture for your referral with ZEB2 in control and ZEB2 knock down cells. We have ~75% knock down in transcript level however in western we do not observe any change.

Detected a major band around 100 kDa (while in theory it should be around 140 kDa). Seems that other people also have similar findings. Is the detected main band truly ZEB2?

Rated 2 out of 5 by from PhDWe got a sample few months ago when we were at VCU (Requester Dongdong Wang). We have tried this antibody using total cell extracts of MDA231 and MCF10A cells with decent results. Although the western blot showed multiples bands.
