Z-DEVD-AFC is a fluorogenic substrate that is used to measure caspase-3 activity in cell lysates. Upon cleavage by caspase-3, the AFC fluorophore is released, resulting in an increase in fluorescence that can be quantified using a fluorometer. This allows monitoring of the activation of caspase-3, a key executioner caspase involved in apoptosis, in response to various stimuli or experimental conditions. The cleavage of Z-DEVD-AFC by caspase-3 occurs at the Asp-Glu-Val-Asp (DEVD) sequence, making it useful for studying the molecular mechanisms of apoptosis.
