Whirlin Antibody (G-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects Whirlin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Whirlin antibody (G-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Whirlin is a cytoplasmic PDZ domain-containing protein crucial for elongation and maintenance of stereocilia, essential mechanosensory organelles found in hair cells of the inner ear. Whirlin co-localizes with actin filaments and is primarily localized in cochlear hair cells, playing a significant role in the dynamic Usher protein interactome. Whirlin interacts with Myosin XVa, a motor protein that binds to the second and third PDZ domains through its C-terminal PDZ-ligand, facilitating transport to stereocilia tips for elongation. The interaction with p55 is critical, as mutations in the DFNB31 gene, which encodes Whirlin, can lead to early loss of p55 labeling in stereocilia, potentially resulting in recessive hearing loss in both rats and humans.
