SEE ALSO...

WASP Antibody (B-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects WASP protein of mouse, rat, human, and mink origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). Anti-WASP antibody (B-9) is raised against amino acids 1-250 of WASP of human origin. WASP (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Wiskott-Aldrich syndrome (WAS) is a genetic disorder linked to a mutation on the short arm of the X chromosome, characterized by symptoms such as thrombocytopenia, eczema, immune system defects, and an increased risk of lymphoproliferative diseases. The gene responsible for WAS encodes WASP, a proline-rich protein that plays a crucial role in cytoskeletal organization. WASP is primarily located in the cytoplasm and is essential for the regulation of actin polymerization, which is vital for various cellular processes, including cell motility, shape, and division. WASP′s location in the cell is underscored by interaction with the Arp2/3 complex, which facilitates the nucleation of new actin filaments, thereby influencing the dynamics of the cytoskeleton. Disruption of WASP function can lead to severe immunological consequences, highlighting WASP′s critical role in maintaining cellular architecture and function.

Can the Antibody WASP (B-9) sc-13139 be used in flow cytometer for human blood analysis? And what is the difference with the antibody WASP D-1sc-5300?

Asked by: giusi85
Thanks for your question. WASP (B-9) is recommended for detection of WASP and N-WASP in human samples by FCM. The difference between the WASP (B-9) and WASP (D-1) is the isotype being mouse IgG2a κ and mouse IgG1 κ respectively. Can this antibody be used in IHC with paraffin-embedded samples?

Asked by: TinTin
Thank you for your question. Yes, this WASP antibody, sc-13139, has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

How can this mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: TinTin
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody, WASP (B-9), is currently available conjugated to agarose (sc-13139 AC), HRP (sc-13139 HRP), phycoerythrin (sc-13139 PE), fluorescein (sc-13139 FITC), Alexa Fluor® 488 (sc-13139 AF488) and Alexa Fluor® 647 (sc-13139 AF647).
