VCX3B encodes VCX-C, a member of the VCX/Y protein family enriched in male germline tissues and associated with testis-specific transcriptional programs. Proteins in this family are implicated in regulating mRNA handling and post-transcriptional control during spermatogenesis, linking VCX-C to processes such as RNA metabolism and germ cell differentiation. VCX3B resides in the X chromosome region that is recurrently involved in copy-number variation, making it relevant for studies of dosage sensitivity and genome stability. Dysregulation of VCX family genes has been explored in the context of male infertility phenotypes and X-linked genomic rearrangements, supporting mechanistic investigations in reproductive and developmental biology models.
VCX-C CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the VCX3B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the VCX3B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the VCX3B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish VCX-C protein expression.
