SEE ALSO...

V-ATPase D1 Antibody (34-Z) is a mouse monoclonal IgG1 antibody that detects V-ATPase D1 in human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). V-ATPase D1, also known as ATP6V0D1, P39, VATX, VMA6, ATP6D, or VPATPD, is a crucial component of the vacuolar-type H+-ATPase complex, which plays a vital role in the acidification of eukaryotic intracellular organelles. V-ATPase D1 is primarily located in the lysosomal membrane, where V-ATPase D1 facilitates the transport of protons across the membrane, maintaining the acidic environment necessary for various cellular processes, including protein degradation and nutrient storage. Proper functioning of V-ATPase D1 is essential for cellular homeostasis, as V-ATPase D1 influences the activity of numerous enzymes and the overall metabolic state of the cell. Dysregulation of V-ATPase activity has been implicated in various diseases, including cancer and neurodegenerative disorders, making anti-V-ATPase D1 antibody (34-Z) a significant tool for therapeutic intervention and research. V-ATPase D1 (34-Z) antibody provides reliable detection for studying the expression and function of V-ATPase D1 in human tissues, contributing to our understanding of its role in health and disease.

I detected only 100kDa band and weak band under 37kDa in HUVECs using this antibody. That's why I am asking about the transfected data in the data sheets.  Can this antibody detect other component of V-ATP ase?

Asked by: Maxi
Thank you for your question. Please contact our Technical Service Department or your local distributor for assistance in troubleshooting this antibody for Western blotting.

The molecular weight of V-ATPase D1 is 40kDa and the WB data of Hela in the data sheet is 40kDa. But WB of human V-ATPase D1 transfected 293T cells in the data sheet is around 100kDa. Which is the true molecular weight?

Asked by: Maxi
Thank you for your question. Thank you for bringing this discrepancy to our attention. The observed molecular weight in the transfected lysate is much higher than expected. We will be reviewing this data for accuracy and will remove or replace it if necessary.

Can sc-81887: V-ATPase D1 (34-Z) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Professor Griffin
Thank you for your question. Yes, sc-81887: V-ATPase D1 (34-Z) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
