USP33 Antibody (1D7) is a mouse monoclonal IgG1 kappa light chain antibody that detects the USP33 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). USP33 (1D7) antibody is available as a non-conjugated antibody. USP33, also known as ubiquitin specific peptidase 33 or VHL-interacting deubiquitinating enzyme 1, is a crucial 942 amino acid protein that plays a significant role in the ubiquitin-proteasome system, which is essential for regulating protein degradation and maintaining cellular homeostasis. USP33 is primarily located in the cytoplasm and nucleus, where USP33 functions as a deubiquitinating enzyme, removing ubiquitin moieties from target proteins and thus preventing their degradation by the proteasome. USP33′s ability to modulate the levels of ubiquitinated proteins is vital for various cellular processes, including signal transduction, transcriptional regulation, and cell cycle progression. Additionally, USP33 interacts with the von Hippel-Lindau (VHL) protein, which is involved in the regulation of hypoxia-inducible factors, further underscoring USP33′s importance in cellular responses to oxygen levels. The expression of three isoforms of USP33, resulting from alternative splicing, adds another layer of complexity to USP33′s regulatory functions, making USP33 a significant target for research in cancer and other diseases where the ubiquitin pathway is disrupted.

Can sc-100632: USP33 (1D7) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jenniferc
Thank you for your question. Yes, sc-100632: USP33 (1D7) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
