uPA Antibody (H77A10) is a mouse monoclonal IgG1 kappa light chain antibody that specifically detects urokinase-type plasminogen activator (uPA), also known as PLAU or urokinase, in mouse, rat, and human samples by western blotting (WB), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). uPA monoclonal antibody (H77A10) was raised against a recombinant protein corresponding to the full-length, glycosylated uPA of mouse origin. uPA is a serine protease crucial for converting plasminogen to plasmin, playing a significant role in fibrinolysis and extracellular matrix degradation. uPA is primarily located on the cell surface, bound to its receptor uPAR, which localizes its proteolytic activity and facilitates processes like cell migration and tissue remodeling. The cell surface localization of uPA allows for targeted proteolysis, essential in wound healing, inflammation, and tumor metastasis. By interacting with uPAR, uPA influences cell signaling pathways that regulate cell adhesion, proliferation, and survival. The activity of uPA is tightly regulated by serine protease inhibitors PAI-1 and PAI-2, which inhibit uPA and control its role in physiological and pathological processes. Anti-uPA antibody (H77A10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental applications.

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody. Rated 1 out of 5 by from No bands seenWestern blotting was performed using twelve different mouse osteosarcoma cell lines which were previously confirmed to express uPA gene based on RT-PCR. Conditions: 12% gel electrophoresis, wet transfer over 2 hours, primary antibodies at 1:200 dilution in 5% mild blocking solution (4C overnight). There was no band visible following the protocol, although positive control (b-actin) was positive.
