Ube2e1 encodes UBE2E1, a ubiquitin-conjugating E2 enzyme that cooperates with E3 ligases to transfer ubiquitin onto substrate proteins, shaping proteostasis and protein turnover. Through its role in ubiquitination, UBE2E1 contributes to regulation of cell-cycle progression, DNA damage responses, and stress-adaptive signaling by modulating the stability and activity of key regulatory proteins. Perturbation of E2 enzyme activity can disrupt ubiquitin-dependent quality control and signaling networks, processes frequently implicated in neurodegeneration, inflammation, and oncogenic transformation. In mouse models, Ube2e1 provides a tractable node for dissecting ubiquitin pathway specificity and redundancy among E2 family members in tissue and disease-relevant contexts.
UBE2E1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ube2e1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ube2e1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ube2e1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish UBE2E1 protein expression.
This CRISPR knockout system enables efficient generation of Ube2e1-deficient cell models for investigation of UBE2E1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
