TSC-22 Antibody (R5-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects TSC-22 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-TSC-22 antibody (R5-6) is available as the non-conjugated format. TSC-22, also known as Transforming Growth Factor Beta-Stimulated Clone-22, plays a crucial role as a transcriptional regulator, influencing cell growth, differentiation, and apoptosis. Notably, TSC-22 contains a leucine zipper domain and a nuclear export signal, which facilitate localization in the cytoplasm under normal conditions. However, during apoptosis, TSC-22 translocates to the nucleus, where TSC-22 exerts transcriptional regulatory functions. This dynamic localization underscores TSC-22′s role in the TGF-beta signaling pathway and the peroxisome proliferator-activated receptor gamma (PPARγ) pathway, both implicated in tumor suppression. Downregulation of TSC-22 mRNA in various cancers, including brain and salivary gland tumors, highlights TSC-22′s potential as a critical player in inhibiting cell proliferation and suggests importance in cancer research and therapeutic strategies.

Hello. I want to know the epitope of TSC-22 to which the sc-101195 antibody binds. Can you tell us the exact location?

Asked by: pico750
Thank you for your question. TSC-22 Antibody (R5-6): sc-101195 has been raised against a recombinant human TSC-22 construct containing the N-terminal amino acids 1-145 of TSC-22. The exact epitope sequence has not been mapped yet.

We are interested in the identification of TSC22D1 expression in mouse fibroblasts and liver samples. The antibody "TSC-22 Ab (R5-6): sc-101195" refers to TSC22D1 or any of the TSC22D1-4 proteins identified so far are reacting with this SCBT antibody?

Asked by: Hagi10
Thank you for your question. TSC-22 Antibody (R5-6): sc-101195 has been raised against recombinant TSC-22 of human origin (encoded by the human TSC22D1 gene). There is little homology between TSC22D1 and TSC22D2-4, so it is unlikely that the antibody recognizes these proteins as well. Please contact our technical service directly for further information or to arrange for as sample of the antibody to test it in your application.

I am using TSC-22 (R5-6): sc-101195 for immunofluorescence, what dilution of this antibody should I use?

Asked by: jenniferc
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
