TRα1/β1 Antibody (C4) is a mouse monoclonal IgG1 kappa light chain antibody that detects the TR alpha 1/beta 1 protein of human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). TRα1/β1 (C4) monoclonal antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Thyroid hormone nuclear receptors (TRs) are crucial ligand-dependent transcription factors that play a significant role in regulating metabolic and developmental processes. TRα1 can localize to both the nucleus and the cytoplasm, and is the only TR imported into the mitochondrial matrix, highlighting its importance in cellular metabolism and energy regulation. This localization allows TRα1 to influence mitochondrial gene expression and function, thereby impacting cellular respiration and energy production. TRs bind to thyroid hormone response elements (TREs) with varying affinities, which are influenced by the presence of 3,5,3′-triiodo-L-thyronine (T3). TR transcriptional activity is further modulated through heterodimerization with retinoid X receptors (RXRα, RXRβ, and RXRγ), which enhances regulatory capabilities. Understanding TRα1 function and localization is essential for elucidating its role in thyroid hormone signaling and implications in metabolic disorders.

When should the TransCruz reagent sc-740 X be used instead of the standard form sc-740?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
