TRα1/α2 Antibody (3C185) is a mouse monoclonal IgG1 antibody that detects TRα1/α2 in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. TRα1/α2 plays a crucial role as a ligand-dependent transcription factor that regulates numerous metabolic and developmental processes, making TRα1/α2 essential for maintaining homeostasis within the body. TRα1 is unique as TRα1 can localize both in the nucleus and cytoplasm, and TRα1 is the only thyroid hormone receptor imported into the mitochondrial matrix, highlighting TRα1′s diverse functional roles. In contrast, TRα2, a C-terminal variant of TRα1, does not bind thyroid hormones and acts as a dominant negative antagonist of thyroid hormone signaling, thereby influencing the overall regulatory mechanisms of thyroid hormone action. The ability of TRs to bind to thyroid hormone response elements with varying affinities, influenced by 3,5,3′-triiodo-L-thyronine (T3), further underscores the complexity of their regulatory functions, which are also modulated through heterodimerization with retinoid X receptors. Thank you for your question. Yes, sc-73213: TRα1/α2 (3C185) is recommended for use in IHC with paraffin-embedded sections.
