
TPX2 Antibody (B-5) is a mouse monoclonal IgM antibody that detects TPX2 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TPX2, or targeting protein for Xklp2, is a crucial microtubule-associated protein that plays a significant role in the assembly of microtubules during cell division, particularly in the M phase of the cell cycle. TPX2 is activated by Ran-GTP, which facilitates chromatin-induced microtubule assembly, ensuring proper spindle formation and function. The structural integrity of TPX2 is vital as TPX2 interacts with Aurora-A kinase at the spindle apparatus, where TPX2 may be regulated through phosphorylation, further emphasizing TPX2′s importance in mitotic spindle assembly. Additionally, TPX2 activates Eg2, a kinase that is essential for microtubule dynamics, by enhancing Eg2′s phosphorylation and activity in a microtubule-dependent manner. Notably, TPX2′s function can be inhibited by binding to importin α, a nuclear import factor, which highlights the regulatory mechanisms that control TPX2′s activity. Disruption of TPX2 expression through RNA interference has been shown to lead to significant defects in microtubule organization during mitosis, underscoring TPX2′s critical role in maintaining cellular integrity during cell division.
