Top3a encodes DNA topoisomerase III alpha (Topo IIIα), a type IA topoisomerase that resolves DNA topological stress and recombination intermediates during replication and repair. In mouse cells, Topo IIIα functions within the BTRR complex with BLM, RMI1, and RMI2 to catalyze dissolution of double Holliday junctions, limiting crossover events and promoting faithful genome maintenance. This activity connects Topo IIIα to homologous recombination, replication fork stability, and sister chromatid segregation pathways. Disruption of TOP3A-associated processes is linked to genome instability phenotypes relevant to studies of cancer biology, developmental defects, and DNA repair disorders in mammalian systems.
Topo IIIα CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Top3a gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Top3a together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Top3a open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Topo IIIα protein expression.
