SEE ALSO...

Tomosyn Antibody (15) is a mouse monoclonal IgG1 kappa light chain antibody that detects Tomosyn protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-Tomosyn antibody (15) is available in both non-conjugated and various conjugated forms, including agarose and horseradish peroxidase (HRP). Tomosyn, also known as STXBP5 (syntaxin binding protein 5), LLGL3, or LGL3, is a 1,151 amino acid protein that plays a crucial role in regulating neurotransmitter release and calcium-dependent exocytosis. Tomosyn is primarily located in the cytoplasm, cell junctions, secretory vesicles, and peripheral membrane, where Tomosyn interacts with Syntaxin 1 and Syntaxin 1B. The presence of 14 WD repeats in Tomosyn′s structure is significant, as these motifs facilitate various cellular functions, including chromatin assembly, cell cycle control, and vesicular trafficking. The WD repeats contribute to the formation of a tertiary propeller structure, which is essential for Tomosyn′s regulatory capabilities. Additionally, Tomosyn inhibits membrane fusion and is involved in the assembly of SNARE complexes, which are critical for the fusion of transport vesicles with the plasma membrane. The existence of multiple isoforms of Tomosyn, resulting from alternative splicing events, further highlights the complexity and versatility of Tomosyn′s function in cellular processes.

I am using Tomosyn (15): sc-136105 for immunofluorescence, what dilution of this antibody should I use?

Asked by: TinTin
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
