TNS2 encodes tensin-2 (Tns2), a focal adhesion–associated cytoskeletal adaptor that links integrin receptors to the actin network and organizes signaling at cell–matrix contact sites. Through its modular domains, Tns2 coordinates assembly and turnover of adhesion complexes and modulates pathways involved in cell spreading, migration, and mechanotransduction, including phosphoinositide-dependent signaling and receptor tyrosine kinase–coupled events. Altered TNS2 function has been associated with dysregulated adhesion signaling and tissue homeostasis, with particular relevance to kidney glomerular biology and podocyte integrity. These properties make TNS2 a useful node for studying adhesion-driven signaling, cytoskeletal remodeling, and context-dependent changes in cellular invasiveness and differentiation.
Tns2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TNS2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TNS2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TNS2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Tns2 protein expression.
This CRISPR knockout system enables efficient generation of TNS2-deficient cell models for investigation of Tns2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
