TIMP-2 Antibody (3A4) is a mouse monoclonal IgG2a kappa light chain antibody that detects TIMP-2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-TIMP-2 antibody (3A4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. TIMP-2 plays a crucial role in regulating extracellular matrix remodeling by inhibiting matrix metalloproteinases, which are enzymes responsible for degrading various components of the extracellular matrix. Such regulation is vital for maintaining tissue homeostasis and is particularly important in processes such as wound healing, tissue repair, and embryonic development. Dysregulation of TIMP-2 can lead to pathological conditions, including fibrosis and cancer metastasis, highlighting TIMP-2′s significance in both normal physiology and disease states. TIMP-2 also interacts with other tissue inhibitors of metalloproteinases, such as TIMP-1, TIMP-3, and TIMP-4, to form complexes with metalloproteinases like collagenases and gelatinases, resulting in irreversible inactivation of these enzymes. This intricate balance between metalloproteinases and their inhibitors is essential for proper cellular function and tissue integrity.

How can TIMP-2 (3A4): sc-21735 monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: jenni
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody, c-Myc (9E10), is currently available conjugated to phycoerythrin (sc-21735 PE), fluorescein (sc-21735 FITC), Alexa Fluor® 488 (sc-21735 AF488) or Alexa Fluor® 647 (sc-21735 AF647)

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with TIMP-2 (3A4): sc-21735?

Asked by: jenni
We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
