TIA-1/TIAR Antibody (D-9) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 21-140 of human TIA-1, reactive with mouse, rat, and human species, and validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TIA-1 and TIAR are closely related RNA-binding proteins that possess three RNA recognition motifs (RRMs) critical for their roles in post-transcriptional regulation of gene expression. These proteins are essential for controlling mRNA stability, translation, and splicing, particularly during stress responses, where they contribute to the formation of stress granules. TIA-1 is primarily located in the cytotoxic granules of lymphocytes, where TIA-1 plays a pivotal role in mediating apoptosis, while TIAR is predominantly nuclear under normal conditions and is expressed in a wide range of cell types, including those of hematopoietic origin. Upon activation of the FAS pathway, TIAR translocates to the cytoplasm, where TIAR participates in apoptosis by promoting the fragmentation of DNA, a hallmark of programmed cell death. Both TIA-1 and TIAR are substrates of FAS-activated serine/threonine kinase (FAST), which is activated in response to FAS ligation. This pathway underscores the importance of TIA-1 and TIAR in regulating immune responses, apoptosis, and cellular adaptation to stress, making them critical targets for research in cancer, autoimmunity, and viral immunity.
