TGase1 Antibody (E-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects TGase1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TGase1 (E-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TGase1, also known as transglutaminase K, plays a crucial role in forming the cross-linked cell envelope (CE) in terminally differentiating mammalian epidermal cells, which is essential for maintaining skin integrity and barrier function. The CE, a 10 to 20 nm thick protein deposit located on the intracellular surface of the plasma membrane, is generated through the action of transglutaminases, which catalyze the formation of isopeptide bonds by cross-linking glutamine and lysine residues in substrate proteins. This process is vital for various biological functions, including epidermal differentiation and the regulation of cell adhesion, which ultimately influences skin health and resilience. TGase1 is a membrane-associated protein consisting of 817 amino acids, and calcium ions regulate TGase1 activity, highlighting TGase1′s importance in cellular signaling and structural integrity. Additionally, TGase1 interacts with other proteins involved in skin barrier formation, further underscoring TGase1′s significance in dermatological research and potential therapeutic applications.

Rated 2 out of 5 by from Not good for WBI tested this antibody for western blotting on whole lysate of HeLa cell line (50 µg) and human keratinocytes (30 µg). I used 1:500 dilution in 2.5% milk dissolved in TBS-0.1%Tween incubated over-night at 4°C. The full-length at 106 kDa was not detected. The antibody seems to recognize only aspecific proteins.
