Testican-2 Antibody (B-5) is a mouse monoclonal IgG2b kappa light chain antibody that detects Testican-2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Testican-2 (B-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Testican family, also known as the BM-40/SPARC/osteonectin family, consists of highly conserved, extracellular, calcium-binding sulfate proteoglycans that play crucial roles in cellular processes. Testican-2 is predominantly expressed in the central nervous system (CNS), with significant presence in regions such as the olfactory bulb, cerebral cortex, thalamus, hippocampus, cerebellum, and medulla, as well as in lung and testis tissues. Notably, Testican-2 is unique among its family members as Testican-2 not only inhibits membrane-type matrix metalloproteinases (MT-MMPs) but also counteracts the inhibitory effects of other Testican family members on pro-MMP-2 activity. This distinctive function is vital for regulating extracellular matrix remodeling and neurite growth, as Testican-2 has been shown to halt neurite extension from cerebellar neurons, suggesting involvement in CNS development and plasticity. The ability of Testican-2 to bind to the COOH-terminal extracellular calcium-binding domain of N-Tes, an amino-terminal splice variant of Testican-3, further underscores its regulatory role in the intricate signaling pathways that govern neuronal development and function.
