TESPA1 (thymocyte-expressed, positive selection-associated 1) encodes an adaptor protein enriched in lymphoid cells that coordinates signal transduction downstream of antigen receptors. In T cells, TESPA1 contributes to proximal TCR signaling and calcium mobilization by linking receptor engagement to PLCγ-dependent pathways and endoplasmic reticulum Ca²⁺ release, thereby shaping activation programs and thymocyte selection. It is also implicated in B cell receptor signaling and broader immune-cell communication networks that influence cytokine responses and differentiation. Altered TESPA1 activity has been associated with immune dysregulation phenotypes, including susceptibility to autoimmune and inflammatory disease mechanisms, making it relevant for studying signaling thresholds and lymphocyte function.
TESPA1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TESPA1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TESPA1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TESPA1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TESPA1 protein expression.
This CRISPR knockout system enables efficient generation of TESPA1-deficient cell models for investigation of TESPA1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
