TCR α Antibody (H28-710) is a hamster monoclonal IgG2b antibody that detects TCR α protein of mouse origin by western blotting (WB) and immunoprecipitation (IP). TCR α Antibody (H28-710) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The T cell antigen receptor (TCR) plays a crucial role in the immune response by recognizing foreign antigens and initiating intracellular signaling that activates T cells, transitioning them from a resting state to an active one. The TCR is a heterodimer composed of either α and β or γ and δ chains, with approximately 95% of circulating T cells expressing the α/β heterodimer, while the remaining 2-5% express the γ/δ heterodimer. TCR functionality is enhanced by association with CD3 chains and the CD4 or CD8 co-receptors, which are essential for effective signal transduction. T helper cells, characterized by CD4 expression, and T cytotoxic cells, marked by CD8 expression, are vital components of the adaptive immune system. CD4 protein, a membrane glycoprotein with four extracellular immunoglobulin-like domains, increases the binding affinity between TCR and class II major histocompatibility complex (MHC) molecules presented by antigen-presenting cells, thereby facilitating a robust immune response.

Does this antibody bind to the intracellular or extracellular domain of TCR α? I tried using it for flow cytometry to stain cell surface TCR α (concentration: 1:100), but all results were negative.
