TCF7L1 encodes the transcription factor TCF-3, a high-mobility group (HMG) DNA-binding protein that functions as a context-dependent regulator of gene expression downstream of canonical Wnt/β-catenin signaling. By partnering with β-catenin or transcriptional corepressors, TCF-3 helps control cell fate decisions, proliferation, and lineage specification programs, including stem and progenitor cell maintenance. TCF7L1 activity intersects with developmental and differentiation pathways and contributes to chromatin-state regulation at Wnt-responsive enhancers. Dysregulated TCF7L1/TCF-3 signaling has been studied in relation to altered differentiation states and proliferative phenotypes in disease-relevant cellular models, including cancer-associated Wnt pathway perturbations.
TCF-3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TCF7L1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TCF7L1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TCF7L1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TCF-3 protein expression.
This CRISPR knockout system enables efficient generation of TCF7L1-deficient cell models for investigation of TCF-3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
