TNFSF13B encodes TALL-1 (also known as BAFF), a TNF superfamily cytokine essential for B cell survival, maturation, and immunoglobulin production. TALL-1 signals through receptors including BAFF-R, TACI, and BCMA to activate NF-κB and related survival pathways, shaping peripheral B cell homeostasis and humoral immune responses. Dysregulated TNFSF13B expression or signaling is associated with altered B cell tolerance and aberrant plasma cell persistence, linking this axis to autoimmune-associated immune activation and B cell–driven inflammatory states. As a soluble and membrane-associated ligand, TALL-1 also influences immune cell crosstalk within lymphoid tissues and inflamed microenvironments.
TALL-1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TNFSF13B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TNFSF13B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TNFSF13B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TALL-1 protein expression.
This CRISPR knockout system enables efficient generation of TNFSF13B-deficient cell models for investigation of TALL-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
