T-type Ca++ CP α1H Antibody (G-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects T-type Ca++ CP alpha 1H protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). T-type Ca++ CP α1H (G-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. T-type Ca++ CP alpha 1H protein, also known as CACNA1H or Cav3.2, plays a crucial role in regulating calcium ion entry into excitable cells, which is essential for numerous physiological processes such as muscle contraction, neurotransmitter release, and gene expression. CACNA1H is primarily located in the plasma membrane of neurons and muscle cells, where CACNA1H contributes to generating low-threshold calcium currents vital for initiating action potentials. The unique biophysical properties of T-type calcium channels allow activation at relatively hyperpolarized membrane potentials, making these channels critical for modulating neuronal excitability and cardiac function. T-type channels are implicated in various pathophysiological conditions, including epilepsy and cardiac arrhythmias, highlighting their significance as potential therapeutic targets.

Can this antibody use in mouse brain? Does it have the quality of distinguishing the channel from membrane or cytoplasm

For clear subcellular localization of your target we recommend using a directly fluorescence-conjugated version of this antibody such as sc-377510 AF488.

Could i use this antibody for rat tissue, i bought it according to your recommend (instead of T-type Ca ++ CP alpha 1H Antibody (H-300))?

We have been using H-300 in our lab. I was wondering if G-10 binds to the same epitope as H-300?

Thank you for your question. Our monoclonal alternative T-type Ca++ CP α1H Antibody (G-10): sc-377510 has been raised against the same epitope region as the discontinued polyclonal H-300 clone, amino acids 2174-2353 of T-type Ca++ CP α1H of human origin.

Can T-type Ca++ CP α1H (G-10): sc-377510 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, T-type Ca++ CP α1H (G-10): sc-377510 has been validated for use in IHC with paraffin-embedded sections. Rated 1 out of 5 by from Immunohistochemistry frozen & free floatingI tested the antibody for an immunohistochemistry on mouse embryo E12.5, fixed 1h in paraformaldehyde 4%. Dilution: 1/50, Permeabilization solution: PBS 1X- Triton 0,2%(& 0,4%) - Goat serum 10%. It did not work.

Was able to recognize all of them, albeit when used as higher concentrations (1:500). Clean bands though with minimal background.
