Surf6 encodes SURF-6, a conserved nucleolar protein implicated in ribosome biogenesis and nucleolar homeostasis through roles in pre-rRNA processing and maturation of ribosomal subunits. SURF-6 participates in RNA-binding and nucleolar organization processes that coordinate protein synthesis capacity with cell growth and proliferation programs. Disruption of nucleolar function and ribosome production is tightly linked to cellular stress responses, altered translation, and proliferation defects, making Surf6 relevant for studies of genome stability and growth control in mammalian systems. In mouse models, Surf6 perturbation provides a tractable entry point to investigate how ribosome biogenesis intersects with developmental phenotypes and disease-associated dysregulation of nucleolar pathways.
SURF-6 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Surf6 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Surf6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Surf6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish SURF-6 protein expression.
This CRISPR knockout system enables efficient generation of Surf6-deficient cell models for investigation of SURF-6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
