Stim1 Antibody (D-2) is a mouse monoclonal IgG2b antibody that detects Stromal Interaction Molecule 1 (Stim1) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Stim1 is a crucial cell surface transmembrane glycoprotein that plays a significant role in mediating calcium influx, particularly following the depletion of intracellular calcium stores. Located primarily in the endoplasmic reticulum, Stim1 functions as a calcium sensor due to its EF-hand domain, which undergoes substantial conformational changes in response to fluctuating calcium levels. When intracellular calcium levels drop, Stim1 translocates to the plasma membrane, where Stim1 activates the calcium release-activated calcium (CRAC) channel subunit, TMEM142A/Orai1, facilitating the influx of calcium ions essential for various cellular processes, including secretion and transcription. Additionally, Stim1′s interaction with Stim2 serves as a regulatory mechanism, as Stim2 inhibits Stim1-mediated store-operated calcium entry, highlighting the delicate balance of calcium signaling in cellular functions. The implications of Stim1 extend to tumor growth suppression and the interactions between stromal and hematopoietic cells, making Stim1 a vital protein in both normal physiology and disease states.
