SEE ALSO...

STI1 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects STI1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). STI1 Antibody (D-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Stress-induced phosphoprotein 1 (STI1) plays a crucial role as a co-chaperone for heat shock proteins HSP70 and HSP90, particularly during cellular heat shock response, which is vital for maintaining protein homeostasis under stress conditions. STI1 is predominantly located in the cytoplasm and can also be found in the nucleus, where STI1 assists in proper folding and stabilization of client proteins, thereby preventing aggregation and misfolding that can lead to cellular dysfunction. STI1′s ability to exist as either a monomer or a dimer enhances functional versatility, allowing STI1 to effectively organize and facilitate interaction between HSP70 and HSP90, which is essential for proper functioning of these chaperones. By acting as a scaffold for client protein recruitment to the relaxed, ADP-bound conformation of HSP90, STI1 suppresses ATP turnover during the loading phase, ensuring that client proteins are correctly processed and folded, which is critical for cellular resilience and response to stress.

Can STI1 (D-6): sc-390203 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Professor Griffin
Thank you for your question. Yes, STI1 (D-6): sc-390203 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Date published: 2016-12-20
Rated 5 out of 5 by from Potent and very specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was strong detection of the ~63 kDa target protein in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested.
