SSEA-1 Antibody (480) is a mouse monoclonal IgM kappa light chain antibody that detects Stage-Specific Embryonic Antigen-1 (SSEA-1) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Anti-SSEA-1 antibody (480) is specifically raised against X-irradiated F9 teratocarcinoma stem cells and targets SSEA-1, also known as CD15, Lewis X antigen, or sialyl Lewis X (sLeX). SSEA-1 is a carbohydrate antigen that plays a pivotal role in cell adhesion, migration, and differentiation during embryonic development, making SSEA-1 essential for proper embryogenesis and leukocyte trafficking. SSEA-1 function is particularly important as SSEA-1 mediates cell-cell interactions through binding to selectins, which are crucial for the movement of leukocytes to sites of inflammation and for the overall development of the immune system. Structurally, SSEA-1 is a tetrasaccharide carbohydrate epitope that influences various cell signaling pathways, thereby impacting cellular behavior. In murine models, SSEA-1 serves as a marker for undifferentiated embryonic stem cells, highlighting SSEA-1′s significance in stem cell research and the understanding of developmental processes. Furthermore, SSEA-1/CD15 is expressed on mature granulocytes and certain cancer cells in humans, linking SSEA-1 to immune responses and cancer metastasis. SSEA-1 (480) monoclonal antibody facilitates the exploration of stem cell biology, cell differentiation pathways, and the mechanisms underlying various diseases.

What exactly does it mean that this antibody is "monoclonal mouse IgM κ (kappa light chain)" antibody? Is there no heavy chain present in this antibody molecule? Can standard anti-mouse IgM secondary antibodies recognize this IgM κ antibody?

Thank you for your question. SSEA-1 (480): sc-21702 is a complete monomeric IgM antibody with heavy and light chains. It can be detected either by regular anti-mouse IgM antibody or by using one of our recommended κ light chain-specific secondary binding proteins, such as m-IgGκ BP-HRP sc-516102, or one of our CruzFluor conjugated κ binding proteins. What secondary antibody may I use for WB? The antibody sc-21702 is a IgM Light chain.

What secondary reagent should I use if I want to perform IF studies in mice tissue samples?

The membrane signal is perfect and there is no background noise. Conditions:   - Demasking with Pt Link from Dako (pH6 or pH9)   - Dilution 1/500 and 1/200 works well   - Secondary antibody used: Alexa anti-mouse 647
