SPT3 Antibody (71-S) is a mouse monoclonal IgM antibody that detects SPT3 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). SPT3 (71-S) antibody is available as the non-conjugated form. SPT3 protein plays a crucial role in the Saccharomyces cerevisiae SAGA complex, a multifunctional coactivator that regulates transcription by RNA polymerase II. Within yeast, SPT3 is integral to the SPT-ADA-GCN5 acetyltransferase (SAGA) complex, which is essential for integrating proteins that possess transcription coactivator and adaptor functions, as well as histone acetyltransferase activity. This complex is vital for core promoter-selective functions, particularly through interactions with the TATA-binding protein. In humans, the STAGA complex, which contains homologs of most yeast SAGA components, exhibits acetyl coenzyme A-dependent transcriptional coactivator functions from chromatin-assembled templates in vitro and associates with spliceosome-associated proteins in HeLa cells. Notably, amino acid sequence comparisons between human SPT3 and yeast counterparts reveal three highly conserved domains, with the most conserved 92-amino acid N-terminal domain showing 25% identity with human TAFII18, underscoring the evolutionary significance and functional conservation of SPT3 in transcriptional regulation.
