


SPA-1 Antibody (3) is a mouse monoclonal IgG1 antibody that detects SPA-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). SPA-1, or signal-induced proliferation-associated gene-1, serves as a crucial RAP1 GTPase-activating protein primarily found in hematopoietic progenitors and peripheral T cells, where anti-SPA-1 antibody (3) plays a significant role in modulating immune responses. SPA-1 is predominantly expressed in fetal and adult lymphohematopoietic tissues, and SPA-1 expression is upregulated in normal lymphocytes following various mitogenic stimuli. By disrupting LFA-1-ICAM1-mediated adhesive interactions, SPA-1 inhibits T cell receptor triggering and interleukin-2 production, potentially through the inhibition of RAP1 activity. Notably, SPA-1 also interacts with aquaporin-2 (AQP2), influencing AQP2 trafficking to the apical membrane, which is essential for maintaining water homeostasis in renal cells. Mice lacking the SPA-1 gene exhibit age-dependent T cell immunodeficiency and develop myeloproliferative disorders that resemble human chronic myeloid leukemia, underscoring SPA-1′s importance in immune regulation and cellular signaling pathways.
