SP140 encodes a nuclear chromatin-associated protein predominantly expressed in hematopoietic and immune lineages, where it contributes to transcriptional regulation and cell state control. As a member of the SP100/SP140 family containing SAND, PHD, and bromodomain-like modules, SP140 is linked to recognition of chromatin features and assembly of regulatory complexes that tune inflammatory and differentiation programs. It has been implicated in innate and adaptive immune signaling pathways, including modulation of cytokine-driven transcriptional responses and macrophage activation states. Genetic and functional studies associate altered SP140 activity with immune-mediated disorders, making it a useful target for dissecting mechanisms of immune gene regulation and epigenetic control.
SP140 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SP140 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SP140 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SP140 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish SP140 protein expression.
This CRISPR knockout system enables efficient generation of SP140-deficient cell models for investigation of SP140 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
