SOAT1 Antibody (ACAT-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects SOAT1 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-SOAT1 antibody (ACAT-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SOAT1, also known as sterol O-acyltransferase-1, is a homotetrameric enzyme that plays a crucial role in lipid metabolism by catalyzing the conversion of cholesterol and long-chain fatty acyl-coenzyme A into cholesterol esters. This enzymatic activity is vital for maintaining cellular cholesterol homeostasis, which is essential for various cellular functions, including membrane integrity and signaling. The gene encoding human SOAT1 is located on chromosome 1 and is predominantly expressed in the endoplasmic reticulum of tissues such as the liver, kidney, adrenal glands, and macrophages. Dysregulation of SOAT1 activity is implicated in the formation of foam cells, a key event in the development of atherosclerosis, making SOAT1 a target for therapeutic intervention. Additionally, SOAT1 interacts with other proteins involved in lipid metabolism, further underscoring SOAT1′s importance in cellular lipid regulation and potential implications in cardiovascular diseases.

I am using SOAT1 (ACAT-1): sc-69836 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

I have tried WB by MIAPaCa2 cell line (said positive control in some reports), but there was no band around 50kDa, and a very intensive band was around 37kDa. What is the band around 37kDa? And please tell me any other positive control cell line.

Thank you for your question. We recommend THP-1 Cell Lysate: sc-2238 (https://www.scbt.com/scbt/product/thp-1-cell-lysate?requestFrom=search) as a positive control for detection of SOAT1 using this mouse monoclonal antibody. We recommend loading 50 µg protein for detection. We do not have any data in MIA PaCa-2 cell line with any of our SOAT1 antibodies, so we can not comment on the expression of the protein in that sample. We have not characterized any bands detected around 37 kDa, however it also appears that we have also observed a similar signal in the published Western blot data on our website. We believe these may be non-specific, since the expected molecular weight for SOAT1 is ~ 50 kDa, although no additional testing has been done.
