Smo Antibody (E-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects Smo protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Smo antibody (E-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Smo protein plays a critical role in the Hedgehog signaling pathway, which regulates cell growth, differentiation, and tissue patterning during embryonic development. Aberrant activation of this pathway, often due to mutations in Smo gene or regulatory components, has been implicated in various cancers, making Smo protein a significant target for therapeutic intervention. Smo (E-5) antibody recognizes a seven transmembrane receptor that transmits signals from Hedgehog ligand to downstream effectors, including GLI transcription factors, thereby influencing gene expression and cellular responses. Understanding Smo protein structure and function is vital, as Smo protein mediates Hedgehog signaling effects and interacts with other pathway proteins, such as Patched, which serves as a negative regulator. The balance between these proteins is crucial for normal development and prevention of tumorigenesis, highlighting Smo protein′s importance in both developmental biology and cancer research.

When I use this antibody in Western blot experiments, I always observe multiple bands. I wonder if this could be related to my denaturation method. I usually add 5× loading buffer to the protein lysate and incubate it at 37°C for 30 minutes. Do you have

Hello, my lab works with S. purpuratus sea urchins. Could you please check if the antibody will be compatible with our species? This is the NCBI reference sequence for the Smo protein in urchins: XP_030830155.1

Has this antibody been tested in Xenopus, either WB or IF?

Thank you for your question. Anti-Smo Antibody (E-5) has only been tested for detection of Smo of mouse, rat and human origin by WB, IP, IF and ELISA. It has not been tested in samples from Xenopus.

Fresh liver samples were homogenized in RIPA buffer, then 40ug of total protein were subjected to 10% gel for SDS-PAGE and transferred onto a PVDF membrane. After blocking with 5% skim milk in 1×TBS-T, membrane were incubated overnight at 4℃ with Smo antibodies(1:1000).And then, it were incubated with anti- mouse IgG(1:2000) at room temperature and detected with ECL kit.
