SmcY Antibody (4C6) is a mouse monoclonal IgG1 kappa light chain antibody that detects SmcY protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-SmcY antibody (4C6) is available as the non-conjugated format. SmcY protein, also known as JARID1D (Jumonji/ARID domain-containing protein 1D), HY, HYA, or KDM5D, is a crucial nuclear protein that belongs to the JARID1 histone demethylase family. Encoded by a gene located on the Y chromosome, SmcY is exclusively expressed in male tissues, where SmcY plays a vital role as a histone demethylase, specifically removing methyl groups from Histone H3. This demethylation is essential for regulating gene expression and maintaining the histone code, which influences chromatin structure and function. SmcY utilizes ascorbate (vitamin C) and iron as cofactors and features several functional domains, including a JMJC domain that is integral to enzymatic activity. Notably, SmcY can be cleaved into a small peptide that acts as a minor histocompatibility antigen, known as HY or HYA, which is implicated in the rejection of female tissue and may play a significant role in male development and fertility. Additionally, the existence of multiple isoforms of SmcY due to alternative splicing events highlights the complexity and regulatory potential of this protein in various biological processes.
