SKAP Antibody (F-11) is a mouse monoclonal IgM antibody that detects SKAP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SKAP (F-11) antibody is available as the non-conjugated anti-SKAP monoclonal isotype antibody. SKAP protein, also known as small kinetochore associated protein or KNSTRN, plays a crucial role in proper alignment and segregation of chromosomes during mitosis, which is vital for maintaining genomic stability. Located in both nucleus and cytoplasm, SKAP is an essential component of mitotic spindle, where SKAP interacts with spindle microtubules to ensure accurate chromosome segregation. Specifically, SKAP forms a complex with SPAG5, localizing to ends of microtubules and stabilizing attachments between microtubules and kinetochores. This stabilization is critical for metaphase-to-anaphase transition, as SKAP promotes normal timing of sister chromatid separation and maintains architecture of spindle poles. Mutations in gene encoding SKAP, situated on human chromosome 15q15.1 and existing as three alternatively spliced isoforms, have been associated with cutaneous squamous cell carcinomas, highlighting SKAP′s significance in cancer biology. Additionally, SKAP expression is regulated by nitric oxide, further underscoring SKAP′s role in cellular signaling pathways.
