Sex hormone–binding globulin (SHBG) is a secreted plasma glycoprotein that binds and transports androgens and estrogens, thereby regulating the free fraction of circulating steroids and modulating hormone availability to target tissues. By buffering steroid distribution and influencing hormone delivery to cells, SHBG contributes to endocrine homeostasis and steroid-responsive transcriptional programs that shape metabolic and reproductive physiology. Variation in SHBG expression or function has been linked to altered sex steroid bioavailability and is frequently studied in relation to metabolic traits and hormone-dependent disease biology, including dysregulated glucose–lipid metabolism and reproductive endocrine phenotypes. In vitro and in vivo models of SHBG perturbation are used to dissect mechanisms connecting steroid transport to downstream signaling networks and gene expression changes in liver and peripheral tissues.
SHBG CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SHBG gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SHBG together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SHBG open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish SHBG protein expression.
This CRISPR knockout system enables efficient generation of SHBG-deficient cell models for investigation of SHBG signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
