SH3D19 Antibody (397.2) is a mouse monoclonal IgG3 antibody that detects SH3D19 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SH3D19, also known as EBP, EEN-binding protein, EVE1, or Kryn, is a 790 amino acid protein that is widely expressed, with the highest levels found in the kidney, heart, lung, liver, skeletal muscle, and small intestine. SH3D19 is primarily localized in the cytoplasm but can be recruited to the nucleus by a fusion protein known as MLL-EEN (mixed-lineage leukemia-endophilin), highlighting a dynamic role in cellular processes. SH3D19 is crucial for regulating the involvement of ADAM (A disintegrin and metalloproteases) proteins in epidermal growth factor receptor (EGFR)-ligand shedding pathways, which are essential for cell signaling and communication. SH3D19 contains five SH3 domains, which facilitate protein-protein interactions, and may function to suppress Ras-induced cellular transformation and the activation of downstream effectors such as Elk-1. Notably, translocation events involving SH3D19 have been implicated in acute myeloid leukemia, suggesting a potential role in carcinogenesis. Additionally, five isoforms of SH3D19 are expressed due to alternative splicing events, further underscoring the complexity and significance in various biological contexts.
