SCARA5 (scavenger receptor class A member 5) is a cell-surface pattern recognition receptor implicated in the binding and internalization of specific ligands, including ferritin, linking it to iron handling and receptor-mediated endocytosis. In human cells, SCARA5 contributes to regulation of cellular adhesion and migration and can influence inflammatory signaling outputs through scavenger receptor–associated processes. Altered SCARA5 expression has been reported across multiple tumor contexts and in fibrotic or inflammatory microenvironments, where it may correlate with changes in epithelial–mesenchymal programs and extracellular matrix remodeling. These features make SCARA5 a useful target for mechanistic studies of iron-associated signaling, membrane trafficking, and microenvironmental regulation of cell behavior.
SCARA5 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SCARA5 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SCARA5 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SCARA5 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish SCARA5 protein expression.
This CRISPR knockout system enables efficient generation of SCARA5-deficient cell models for investigation of SCARA5 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
