RUFY1 Antibody (SS-41) is a mouse monoclonal IgG2a antibody that detects RUFY1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). RUFY1, also known as RUN and FYVE domain containing 1, RABIP4, or ZFYVE12, is a 708 amino acid protein primarily localized to the cytoplasm and early endosome membrane, playing a crucial role in early endosomal trafficking by binding to phosphatidylinositol 3-phosphate-containing vesicles. This interaction is vital for the proper sorting and transport of proteins within the cell, which is essential for maintaining cellular homeostasis and responding to environmental changes. RUFY1 features one RUN domain and one FYVE-type zinc finger, which facilitate binding to phosphatidylinositol 3-phosphate, and shows high expression in tissues such as the testis, lung, brain, and kidney. Notably, RUFY1 undergoes post-translational modifications, including phosphorylation by ATM or ATR in response to DNA damage, with specific phosphorylation of Tyr 389 and/or Tyr 400 being necessary for endosomal localization. Additionally, three isoforms of RUFY1 arise from alternative splicing, contributing to functional diversity and regulatory mechanisms within the cell.
