Rictor Antibody (H-11) is a mouse monoclonal IgG2b kappa light chain antibody that detects Rictor protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Rictor (H-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rictor, also known as rapamycin-insensitive companion of FRAP, is a critical component of mTOR signaling pathway, which regulates cell growth, proliferation, and survival in response to nutrients and growth factors. Rictor′s ability to form a complex with FRAP is essential for integrating these signals, and Rictor plays a pivotal role in phosphorylation of Akt/PKB, a key player in cell survival and metabolism. Notably, Rictor directly phosphorylates Akt/PKB on Ser473, which is crucial for full activation, and Rictor also facilitates Thr308 phosphorylation by PDK1. This regulatory mechanism is particularly important in cancer context, as Rictor-FRAP complex may serve as a potential drug target in tumors that have lost expression of PTEN, a tumor suppressor that normally inhibits Akt/PKB activation. The diverse applications of anti-Rictor antibody (H-11) make this tool invaluable for researchers studying intricate pathways of cell signaling and their implications in various diseases.

What do you recommend for Antibody dilution? Nonfat Milk or BSA?

Thank you for your question. The recommended starting dilution for Western blotting is 1:200 (range 1:100-1:1,000). We recommend using 5% nonfat milk in TBST as blocking buffer, and antibody diluent. Please refer to our protocol online for further details.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with Rictor (H-11): sc-271081?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
