RelA Antibody (5G8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the RelA protein of Escherichia coli origin by western blotting (WB). Anti-RelA antibody (5G8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. RelA plays a crucial role in the stringent response, a vital adaptive mechanism that bacteria, such as Escherichia coli, employ during periods of amino acid starvation. This response is characterized by the rapid accumulation of guanosine 3′,5′-bispyrophosphate (ppGpp), a signaling molecule that inhibits RNA synthesis, thereby reducing translation rates and facilitating the conservation of resources within the cell. RelA acts as a ppGpp synthetase, becoming activated when amino acid levels are low, and catalyzes the ATP-dependent synthesis of ppGpp. The activation of RelA occurs when uncharged tRNA molecules enter the A site of the ribosome, signaling a shortage of amino acids and prompting the synthesis of ppGpp to slow down protein production. Once amino acid levels are restored, RelA is hydrolyzed, leading to a decrease in ppGpp synthesis and the resumption of normal translation rates. This regulatory mechanism is essential for bacterial survival and adaptation to fluctuating nutrient conditions, making RelA a significant target for studies related to bacterial physiology and stress responses.
