Ranbp2 encodes Ran BP-2 (Nup358), a major cytoplasmic filament component of the nuclear pore complex that coordinates Ran GTPase–dependent nucleocytoplasmic transport and contributes to nuclear envelope homeostasis. As a SUMO E3 ligase, it regulates SUMOylation of select cargoes and participates in processes including mitotic progression, microtubule dynamics, and proteostasis. Ran BP-2 also interfaces with pathways governing mRNA export and stress responses, linking nuclear transport to cell-cycle control and cellular adaptation. Dysregulation of nucleocytoplasmic trafficking and SUMO-dependent regulation has been associated with neurodegenerative phenotypes and altered immune signaling, making Ranbp2 a useful node for mechanistic studies in mouse systems.
Ran BP-2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ranbp2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ranbp2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ranbp2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Ran BP-2 protein expression.
This CRISPR knockout system enables efficient generation of Ranbp2-deficient cell models for investigation of Ran BP-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
