    Overview

    Rragb encodes Rag B, a Ras-related small GTPase that forms heterodimers with RagA/RagC/RagD to couple intracellular amino acid availability to mTORC1 activation at the lysosomal surface. By regulating recruitment of mTORC1 to the Ragulator complex and coordinating with nutrient-sensing inputs, Rag B influences protein synthesis, autophagy, lysosome function, and metabolic adaptation. Perturbation of Rag GTPase signaling can remodel anabolic–catabolic balance and stress responses, processes frequently implicated in oncogenic growth programs and neurodegenerative and immunometabolic phenotypes. In mouse systems, Rragb is therefore a useful node for dissecting nutrient-driven signaling networks and their downstream transcriptional and translational outputs.

    Rag B CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Rragb gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Rragb together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Rragb open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Rag B protein expression.

    This CRISPR knockout system enables efficient generation of Rragb-deficient cell models for investigation of Rag B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
