Rab11A Antibody (A-6) is a mouse monoclonal IgG2b kappa light chain antibody raised against amino acids 130-216 of human Rab11A, detecting the protein in mouse, rat, and human origins via western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA); anti-Rab11A antibody (A-6) also reacts with additional species including equine, canine, bovine, and porcine. Rab11A, also known as Ras-related protein Rab-11A or RAB11, is a member of the Rab family of small GTPases that regulate intracellular vesicle trafficking, particularly in the recycling endosome pathway. Post-translationally modified by prenylation through geranylgeranylation at C-terminal cysteine residues, Rab11A requires this lipid modification for proper membrane association and function in vesicle formation, movement, and fusion. These post-translational modifications are crucial as they facilitate Rab11A′s localization to specific membrane compartments, influencing essential cellular processes such as receptor recycling, cytokinesis, and maintenance of cell polarity. Dysregulation or mutations affecting Rab11A or its post-translational modifications can lead to disruptions in cellular trafficking pathways, contributing to various diseases. By targeting a specific epitope within Rab11A, Rab11A (A-6) monoclonal antibody provides a valuable tool for researchers studying vesicular transport mechanisms and the role of Rab11A in health and disease. The broad species reactivity and applicability of Rab11A (A-6) antibody across multiple detection methods make this reagent essential for advancing cell biology research and understanding disorders associated with vesicle trafficking dysfunctions.

Hi every one,  I blocked membrane with 5% milk then inucubated with 1:500 Rab11a (sc-166912), the band is faint and there is moe background? Could any body tell me what I should use 5% milk or 2%BSA, It seems to me the problem in blocking buffer

Thank you for your question. We recommend our UltraCruz® Blocking Reagent: sc-516214 which has been optimized for use with all of our antibody products. Generally, BSA is better for revealing faint signals, whereas milk-based blocking buffers can reduce background from promiscuous binding. You may also need to increase the amount of primary and/or secondary antibody in your experiment for a cleaner and more robust signal. Does Rab 11A Antibody (A-6): sc-166912 only recognize Rab11A? Or can the antibody also bind to Rab11B?

Thank you for your question. This monoclonal antibody is raised against an antigen that is specific for RAB 11A and does not cross react with Rab 11B. However, we have not tested the antibody for cross reactivity with Rab 11B specifically.

I have been trying to do immuno-fluorescence with the antibody using a standard Immunostaing protocol. I have tried dilution of 1:10 also, but not got any staining. The antibody gives a good band in western blot . Please kindly suggest.

What is the recommended fixation method for IF with Rab 11A (A-6): sc-166912 monoclonal antibody?
